The english sentence is actually the other way around:
Se joka puhuu vähiten tietää eniten.
I see the difference as rather subtle, like between “He knows most who speaks least” and “He who knows most speaks least”.

Yes, the difference is subtle, but it’s there.
Both mean that knowing much results in talking less.
But the implication is reversed:
He who knows most speaks least:
→ If you know much, you don’t speak much.
He knows most who speaks least:
→ If someone does not speak much, they must be wise.
→ You recognize the wise people by looking at how little they say.
