Why not “Hebe deine Hand, wenn du eine Frage hast.”?
It is simply more idiomatic to use “sich melden” instead of “die Hand heben” when in a classroom context - or a university lecture, it seems, because of the formal you (a professor usually does not use “du” to address the students).
