Ann has no sister.

Ann non ha sorelle → Ann has no sister
Sister without “s” is correct?

You could use both however without the “s” sounds more literary than with an “s”. Or in other words, in everyday speech you would say “sisters”.

Or even, 1) Ann has two brothers but she has no sisters. 2) Ann has a brother but no sister.

Ahia, I did not notice the “e”! Now I understand your query. Hopefully our examples have helped with the “s” problem.
